513 Understanding the Role of HMGB1 Post-Traumatic Brain Injury - The Complex Interplay Between Neuro-Inflammation and Neurogenesis

Abstract Introduction Traumatic brain injury (TBI) is a global public health burden. Although neurogenesis occurs post-injury, achieving long term survival of newly generated neurons remains elusive. High Mobility Group Box protein 1 (HMGB1) pivotal cytokine in hosting the neuro-inflammatory response to injury, but also mediates during physiological development. In this review, we examine current evidence for post-traumatic and HMGB1 as therapeutic target. Method PubMed database was evaluated with following search terms: HMGB1, isoforms, neurogenesis, traumatic Toll-like receptor, receptor advanced glycation end-products. Results Multiple studies support existence post-injury both vitro vivo. Different target receptors mediate different functions though these are not mutually exclusive context injury. Interaction RAGE responsible developmental whilst TLR-4 innate immune response. Though isoforms recognised, specific effects unexplored. vivo animal demonstrate positive antagonism post-TBI, long-term outcomes remain unclear. Conclusions Modulating may enhance post-TBI recovery, mechanistic understanding its on fundamental avoid negating potentially beneficial effects.
